大蒜油联合5-FU诱导腺样囊性癌细胞株ACC-M凋亡的研究

Study on garlic oil combined with 5-FU induced apoptosis of adenoid cystic carcinoma cell line ACC-M

  • 目的:探讨大蒜油联合5-FU诱导腺样囊性癌细胞株ACC-M凋亡的效应。方法:人涎腺腺样囊性癌细胞株ACC-M细胞培养,分为实验组(5-FU组、大蒜油组、大蒜油+5-FU组)和对照组,采用MTT法观察肿瘤细胞的体外生长活性;采用流式细胞术分析肿瘤细胞周期和凋亡率的变化。结果:MTT实验结果显示不同浓度的5-FU、大蒜油、大蒜油+5-FU对ACC-M细胞均有抑制作用,随着浓度和作用时间的增加而呈上升趋势;流式细胞术分析显示细胞周期发生明显变化,随着浓度增加、作用时间延长G0/G1期的细胞比率上升,S期无明显变化,而G2/M期的细胞下降,凋亡率显示大蒜油联合5-FU诱导ACC-M细胞凋亡明显强于单独组。结论:大蒜油能有效诱导腺样囊性癌细胞株ACC-M凋亡,大蒜油联合5-FU应用对ACC-M细胞的作用明显强于大蒜油,5-FU可单独应用。
