长期抗原刺激鼻致敏豚鼠模型的鼻黏膜细胞外基质变化

Changes of extracellular matrix of nasal mucosa in guinea pig model of prolonged allergic-induced rhinitis

  • 目的: 观察抗原长期刺激变应性鼻炎豚鼠模型鼻黏膜细胞外基质(ECM)变化的特征。方法: Hartley豚鼠32只,随机分为4组(每组8只),即实验2 w组、实验6 w组、实验12 w组和对照组。卵清蛋白制备变应性鼻炎动物模型,抗原刺激分别持续2 w、6 w和12 w。在最后一次激发24 h后处死动物取鼻黏膜组织。鼻黏膜组织切片,采用苏木精-伊红染色计数嗜酸粒细胞数量、测量鼻黏膜上皮厚度;三色胶原染色法染色测定ECM胶原蓝染面积百分比;免疫组织化学法检测鼻黏膜组织转移生长因子-β1(TGF-β1)及Ⅰ、Ⅲ型胶原的吸收度。结果: ①各实验组鼻黏膜均有大量嗜酸粒细胞浸润,上皮增厚,与对照组比较差异有统计学意义。②实验2 w组、实验6 w组、实验12 w组和对照组鼻黏膜ECM胶原蓝染面积百分比分别为(18.6±12.0)%、(22.9±9.3)%、(21.7±1.3)%和(15.6±4.0)%;实验6 w组、实验12 w组ECM胶原蓝染面积百分比与对照组比较均差异有统计学意义(均P<0.01),但实验2 w组与对照组比较差异无统计学意义。③同对照组比较,各实验组TGF-β1吸收度增加,差异有统计学意义;Ⅰ、Ⅲ型胶原吸收度在各实验组都呈增加趋势。结论: 随着抗原刺激延长,鼻黏膜慢性炎症导致与重塑相关的炎性因子表达增强,ECM胶原沉积明显。
