Expression and clinical significance of Nrf2 and ERK pathway in papillary thyroid carcinoma

  • 目的:探讨核因子E2相关因子2(Nrf2)及ERK信号通路在甲状腺乳头状癌(PTC)中的表达及临床意义。方法:免疫组织化学法检测PTC组织芯片中Nrf2、ERK2和p-ERK1/2的表达;Western blot检测Nrf2、ERK2、p-ERK1/2蛋白在PTC、癌旁组织及正常甲状腺组织中的相对含量;实时定量PCR检测Nrf2 mRNA在PTC、癌旁组织及正常甲状腺组织中的转录水平。结果:Nrf2、ERK2、p-ERK1/2蛋白在PTC中高表达,在癌旁组织中弱阳性或阴性表达(P<0.01)。Nrf2、ERK2、p-ERK1/2蛋白在淋巴结转移和临床Ⅲ~Ⅳ期组明显高于无淋巴结转移组和临床Ⅰ~Ⅱ期组(P<0.05),且Nrf2蛋白与ERK2和p-ERK1/2蛋白呈正相关(r=0.723 2和r=0.668 6,P<0.05)。Western blot结果显示PTC组织Nrf2、ERK2、p-ERK1/2含量高于癌旁和正常组织(P<0.01)。PTC组Nrf2 mRNA转录水平明显高于癌旁及正常组织(P<0.01)。结论:Nrf2与PTC的进展有关,推测Nrf2高表达与ERK2通路有关。
