木黄酮对人喉癌细胞Hep-2诱导凋亡及抑制增殖的研究

  • 目的:探讨木黄酮对人喉癌细胞Hep-2的诱导凋亡及抑制作用。方法:采用CCK-8法测定木黄酮抑制细胞系Hep-2增殖的IC50值;应用流式细胞术检测木黄酮对喉癌细胞所引起的周期分布变化与凋亡率;倒置相差显微镜下观察木黄酮干预人喉癌细胞Hep-2前后的细胞形态学变化。结果:木黄酮抑制Hep-2细胞增殖的IC50值为23.64 μg/ml;采用浓度为12 μg/ml和24 μg/ml的木黄酮分别作用于喉癌细胞24 h的凋亡率为22.4%±1.65%和30.64%±2.95%,与对照组比较差异均有统计学意义(P<0.05);采用浓度为12 μg/ml和24 μg/ml的木黄酮分别作用于喉癌细胞48 h的凋亡率为30.55%±0.72%和48.69%±1.06%,与对照组比较差异均有统计学意义(P<0.05)。同一浓度的木黄酮作用于喉癌细胞24、48 h的凋亡率差异有统计学意义(P<0.05)。结论:木黄酮能够明显抑制人喉癌细胞Hep-2细胞增殖,并引起Hep-2细胞的凋亡,凋亡率随时间的延长和剂量的增加而增高。
