Expression of LC3 and ECP in allergic rhinitis and their significance

  • 目的:探讨自噬相关基因微管相关蛋白1轻链3(LC3)在变应性鼻炎(AR)患者鼻黏膜中的表达及其与嗜酸粒细胞阳离子蛋白(ECP)的关系。方法:选取20例AR患者下鼻甲黏膜(AR组)和20例正常鼻腔黏膜(对照组),苏木精-伊红染色观察各组鼻黏膜的中嗜酸粒细胞的分布与表达,采用免疫组织化学方法、蛋白免疫印迹法检测各组鼻黏膜中LC3和ECP的表达情况。结果:苏木精-伊红染色法显示在AR组患者鼻腔黏膜中可观察到大量嗜酸粒细胞及淋巴细胞浸润。免疫组织化学结果显示LC3与ECP主要分布于AR鼻黏膜上皮内,较对照组明显增多,差异有统计学意义(P<0.05)。蛋白免疫印迹法示LC3和ECP在AR组中的表达较对照组显著上升(P<0.05),AR组中LC3与ECP的表达呈正相关(P<0.05)。结论:LC3和ECP在AR患者中表达升高且具有相关性,提示自噬在AR的毒性作用中起了重要作用,可为AR的精准化治疗提供理论依据。
