The effect of the nasal packing material on nasal epithelial cells

  • 目的: 探究聚乙二醇双丙烯酸酯复合物水凝胶降解物质对鼻黏膜纤毛传输系统的影响。方法:实验根据不同配比将聚乙二醇双丙烯酸酯、壳聚糖、聚乙烯醇材料通过光交联形成复合物水凝胶,根据国际通行医疗器械生物学评价标准将聚乙二醇双丙烯酸酯复合物水凝胶材料、明胶海绵材料、壳聚糖材料配制成浸提液。实验分为聚乙二醇双丙烯酸酯复合物水凝胶材料组(A组)、明胶海绵材料组(B组)、壳聚糖材料组(C组)及对照组(D组),因各填塞材料用DMEM/F12:BEGM培养液浸提,故选用DMEM/F12:BEGM培养液作对照。取人鼻腔钩突组织,经气液界面培养。高速数字显微视频成像系统检测纤毛摆动频率,分别测量加药前基础状态(0 min)及加药后1 h、1 d、2 d、3 d内的CBF。结果: ①A、B、C、D组随着天数增加,纤毛摆动频率均呈现先增加后减慢的趋势。②A组与其他各组相比,纤毛摆动频率数值变化最大(P<0.05),提示聚乙二醇双丙烯酸酯复合物水凝胶材料可使纤毛摆动频率增加。③B、C、D组间的最大增加量与最大减少量相比,差异无统计学意义(P>0.05),提示可吸收性明胶海绵材料和可吸收性壳聚糖材料对鼻黏膜纤毛活性无影响。结论: 聚乙二醇双丙烯酸酯复合物水凝胶材料可使体外气液界面培养的鼻黏膜上皮细胞纤毛摆动频率增加,从而使鼻黏膜纤毛活性增强,因此可以满足该新材料临床应用的安全性评价需求。
