The function of regulator cAxin2 in inner ear development of chick embry

  • 目的:探讨Wnt家族副调节因子cAxin2对鸡胚内耳早期发育的调控机制。方法:采用增强的绿荧光蛋白、cAxin2和短发夹RNA构建了一个带EGFP的干扰质粒,通过活体电穿孔技术转染在耳囊中,用原位杂交技术检测基因cAxin2、cPax2(Paired Box2)及cBMP4(Bone Morphogenetic Protein 4)在转染后耳囊中的表达。结果:转染侧耳囊显示cAxin2的表达被短发夹RNA抑制,cPax2被下调而cBMP4被上调,并能看到一个异常增大的耳囊。结论:cAxin2 是一个内耳发育过程中非常重要的调控基因。
