Association between IL-27 gene polymorphisms and susceptibility to allergic rhinitis

摘要: 目的:讨论IL-27基因多态性与中国人群变应性鼻炎(AR)易感性是否存在相关性。方法:收集445例AR患者及691例对照组的抗凝血液标本进行病例-对照研究。提取血液样本中的DNA,并运用PCR-RFLP和直接测序的方法对DNA样本中IL-27基因的3个多态位点(rs153109、rs17855750及rs181206)进行基因分型。结果:AR组及对照组IL-27基因的3个SNP分型结果均符合哈迪温伯格平衡(P>0.05)。AR组中rs153109 TT基因型及T等位基因频率显著高于对照组,CC、CT基因型及C等位基因频率显著低于对照组(P<0.05)。rs153109位点TT基因型及T等位基因与AR的易感性显著相关,而CC、CT基因型及C等位基因则显著降低AR的发生率。对于rs17855750位点,TT基因型及T等位基因为易感因素,GT基因型及G等位基因为保护性因素。IL-27基因的TTT及TTC单体型与AR的发生呈正相关,而CGT、CTC 及CTT单体型则降低AR的发生风险。结论: IL-27的rs153109 及rs17855750位点的多态性与AR的易感性有关,该研究为中国汉族人AR易感性的基因学诊断提供了新的理论依据。Abstract: Objective: To evaluate the potential association of IL-27 polymorphisms with allergic rhinitis(AR) in a Chinese Han population.Method: A case-control study was performed in 445 Chinese AR patients and 691 healthy controls. Three SNPs in the IL-27p28 gene, including rs153109, rs17855750 and rs181206, were detected using a polymerase chain reaction-restriction fragment length polymorphism assay(PCR-RFLP).Result: A significantly increased prevalence of the rs153109 TT genotype and the T allele was found in AR patients, while a decreased prevalence of the CT and CC genotypes and the C allele was found. For rs153109, the TT genotype and the T allele were significantly associated with the risk of AR, but the CT and CC genotypes and the C allele decreased the risk of AR; for rs17855750, the TT genotype and T allele were risk factors for AR, and the GT genotype and G allele provided protection. TTT and TTC haplotypes in the IL-27 p28 gene were positively correlated with AR, while CGT, CTC and CTT haplotypes were associated with a significantly decreased risk of AR.Conclusion: This study indicates that IL-27 p28 polymorphisms rs153109 and rs17855750 are likely involved in AR susceptibility, making them potentially useful genetic biomarkers for AR susceptibility in the Chinese Han population.
