Expression of AMCase mRNA in nasal mucosa of allergic rhinitis in SD rats

  • 目的:研究变应性鼻炎(AR)大鼠鼻黏膜AMCase mRNA表达,探讨其与AR的关系。方法:30只SD大鼠,随机分为实验组和对照组。实验组20只,卵清白蛋白(OVA)隔日腹腔注射基础致敏2周,继之每日鼻腔局部激发1周,建立大鼠AR模型;对照组10只,生理盐水代替OVA进行实验。末次激发后根据鼻部症状累积评分评判AR造模是否成功,并取所有大鼠双侧鼻腔及中隔黏膜,常规苏木精-伊红染色病理明确AR诊断。提取鼻黏膜RNA,应用反转录合成cDNA,采用实时荧光定量 PCR(RT-PCR)检测AMCase mRNA在大鼠AR鼻黏膜中的表达。结果:RT-PCR 检测AMCase mRNA表达,显示实验组与对照组差异有统计学意义(P<0.05);②AR组AMCase mRNA表达同鼻部症状评分正相关(r=0.411,P<0.05)。结论:大鼠AR鼻黏膜AMCase mRNA表达量增多,提示AMCase表达上调在变应性鼻炎的发病机制中起作用,抑制该酶活性可以成为AR治疗新的靶点。
