Research of expression of L-DOPA decarboxylase in laryngeal cancer

  • 目的:分析L-多巴脱羧酶(DDC)mRNA和蛋白在喉癌组织中的表达,以评估其在喉癌诊断和预后中的临床意义。方法:从53例喉癌患者的肿瘤切除手术中获取106份组织样品,并从这些组织样品中分别分离出总RNA。采用RT-PCR法对DDC mRNA的表达水平进行量化,同时采用Western Blot蛋白质印迹法对DDC蛋白的表达水平进行检测。结果:与正常喉黏膜组织相比,原发性喉癌(P=0.000)和复发性喉癌(P=0.033)样品中的DDC mRNA表达水平都出现了显著下调。且DDC mRNA表达水平与患者年龄和组织学分级无关,但与TNM分期之间存在显著的负相关性(P=0.021)。同时,我们在肿瘤和正常喉组织中都检测到了DDC蛋白的表达。结论:DDC的表达状态可能在喉癌的发展中发挥重要的作用,在未来的喉癌患者临床管理中具有重要的意义。
