喉癌病变过程中细胞周期调控因子的流式检测及意义

Expression and significance of the cell cycle regulators in laryngeal carcinogenesis detected by flow cytometry

  • 目的:探讨细胞周期调控基因cyclin E、cyclin D1、p21、p16在喉癌发生、发展中的作用。方法:选取23例声带息肉、69例喉癌前病变、33例喉癌石蜡包埋组织为研究对象,切片脱蜡后制备成细胞悬液,采用流式细胞术检测cyclin E、cyclin D1、p21、p16基因的荧光表达情况。结果:在声带息肉、喉癌前病变、喉癌组织中,cyclin E的阳性表达率分别为13.04%、20.29%和42.42%;cyclin D1的阳性表达率分别为26.09%、43.48%和93.94%;p16的阳性表达率分别为61.90%、40.98%和14.28%;p21的阳性表达率分别为47.62%、23.81%和26.23%。提示cyclin D1、cyclin E的阳性率表达形成声带息肉→喉癌前病变→喉癌组织阳性强度逐渐增强的梯度现象,而p21、p16的阳性表达强度则呈逐渐减弱的现象。结论:①细胞周期调控因子的异常表达是喉癌发生的分子事件;②正性调控因子cyclin D1、cyclin E高表达,负性调控因子p16、p21低表达,即多个与细胞周期相关的正负调控因子失衡在喉癌的发生中可能起着重要作用。
