Transplanting neural stem cells in nerve conduit to promote rats facial nerve regeneration

摘要: 目的:观察移植神经干细胞到可降解的聚乳酸/聚羟基乙酸(PLGA)神经导管修复大鼠面神经缺损的可行性。方法:20只SD大鼠随机分为2组,每组进行右侧面神经损伤手术,分别移植PLGA导管(A组)或含有神经干细胞的PLGA管(B组)。术后4、12周分别通过电生理检测、轴突形态学检测等方法来评估面神经的修复效果。结果:术后4周和12周,B组在神经动作电位的振幅分别为(27.53±7.48)、(141.23±21.18)μV,轴突的面积为(10 539.48±3 605.35)、(30 076.22±7 614.05)μm2,都高于A组[术后4周和12周,动作电位振幅为(13.03±11.38)、(118.50±22.70)μV; 轴突面积为(6 633.20±3 759.30)、(20 772.53±10 903.65)μm2](P<0.05)。结论:神经干细胞移植可能会促进面神经的再生。Abstract: Objective: To construct a biodegradable PLGA nerve conduit (NC) filled with NSCs in order to improve facial nerve regeneration. Method: SD rats were subjected to right facial nerve transection, and PLGA NCs filled with NSCs were used to bridge the nerve gap. Facial nerve regeneration was assessed 4 and 12 weeks after surgery, through electrophysiological testing, and morphometric analysis of axons. Result: Nerve action potential amplitude, and axonal area were significantly greater in the NSCs group than the empty NC group. Conclusion: NSCs transplantation may improve regeneration of the facial nerve.
